Zusammenfassung
In 86 Serumproben wurden folgende Bestimmungen durchgeführt: Serumeisen und ungesättigte Eisenbindungskapazität (UEBK), jeweils als Doppelbestimmung mit den Methoden vonSchade [20, 21]; Transferrin mit drei immunologischen Methoden: Agardiffusion nachOudin [9], Immunokrit [8] und Nephelometrie nachSchultze undSchwick [23]. Von jeder Methode wurde durch 20-bzw. 14fache Bestimmung ein und desselben Serums die mittlere Abweichung vom Mittelwert σ ermittelt. Es ergaben sich folgende σ-Werte: Serumeisen: 1,4 γ-%, UEBK: 14 γ-%, Agardiffusion: 9 mg-%, Imunokrit: 10 mg-%, Nephelometrie: 14 mg-%. Die aus dem Serumeisen und der UEBK ermittelte totale Eisen-Bindungs-Kapazität (TEBK) wird auf die entsprechende Transferrinmenge umgerechnet, und die Werte werden statistisch mit den Ergebnissen der immunologischen Bestimmungen verglichen. Die immunologischen Werte liegen alle bedeutend höher als die über die TEBK gewonnenen. Es ergibt sich jedoch eine konstante Korrelation, die beim Vergleich mit der Agardiffusion am ausgeprägtesten ist. Es gilt folgende Umrechnungsformel: Transferrinkonzentration über TEBK=0,598 · Transferrinkonzentration über Agardiffusion + 7. Wir empfehlen zur Bestimmung des Transferrins und der TEBK nach Berücksichtigung aller praktisch wichtigen Faktoren die Agardiffusionsmethode nachOudin.
Summary
The following determinations were performed in 86 serum samples: Serum-iron and unsaturated iron-binding-capacity (UIBC), in double determinations (Schade [20, 21]). Transferrin by 3 immunological methods: 1. Agar gel diffusion (Oudin [9]), 2. Immunocrit [8], 3. Turbidity test (Schultze andSchwick [23]). The reproducibility of each method was estimated by 20 (14) determinations of the same serum sample. We found the following σ values: Serum iron: 1,4 γ-%; UIBC: 14 γ-%; agar gel diffusion: 9 mg-%; immunocrit: 10 mg-%; turbidity test; 14 mg-%. The total iron-binding-capacity (TIBC)=serum iron + UIBC corresponds to calculated amounts of transferrin, which are compared with the results of the immunological determinations. The immunological values are consistently higher than those obtained by the TIBC. There is however a constant correlation, which is the closest between the agar gel diffusion technique and the TIBC. The following conversion formula was calculated: Transferrin concentration by TIBC=0,598 · transferrin concentration by agar gel diffusion + 7. Considering all the factors involved we recommend for the quantitative routine estimation of transferrin and TIBC the agar gel diffusion technique according toOudin.
