The 4-Quinolones: Anti Bacterial Agents in Vitro

Abstract

Varying the inoculum size by 10 000 fold has negligible effects on the minimum inhibitory concentrations (MICs) of 4-quinolones against Escherichia coli (Chin and Neu 1983; Smith 1984). MIC studies, however, only provide a measure of the inhibition of bacterial multiplication by 4-quinolones i.e. only provide information regarding the bacteriostatic activity of these drugs. The bacteriostatic and bactericidal activities of the 4-quinolones are quite distinct. For example, urinary magnesium concentrations have been shown to severely antagonise the bactericidal activity of 4-quinolones but have little effect on their bacteriostatic activity (Ratcliffe and Smith 1983). Conversely, low pH antagonises the bacteriostatic activity of ciprofloxacin more than its bactericidal activity (Ratcliffe and Smith 1985). Therefore it was decided to investigate the effect of initial inoculum size on the bactericidal activity of ofloxacin and ciprofloxacin on E. coli KL16 and Staphylococcus aureus E3T.
