Abstract

Protein synthesis is an important regulatory step in gene expression. The rate of protein synthesis is controlled by the amount of mRNA available to the translational machinery and the translational efficiency of the mRNA. The former involves gene transcription, mRNA processing, mRNA transport to the cytoplasm, and the half-life of the mRNA. Translational efficiency is determined by mRNA structure, the level and activity of the translational machinery, and by factors that influence initiation and elongation rates. Most translational control is thought to be exerted at the level of initiation of protein synthesis. Initiation is controlled primarily by structure at the 5’ end of the mRNA and the activity of translation initiation factors. Factors which control translation initiation may influence essentially all cellular mRNAs or may be mRNA specific.
