Analysis of the Reaction Kinetics of Single Esterasesepharose Beads by Microfluorometry

Abstract

The evaluation of immobilized enzyme kinetics by conventional techniques often relies on batch reactor experiments where equations are derived from hypothetical properties of single beads (1). Batch reactor experiments however can provide only indirect evidence for these hypothetical properties. For direct evidence methods have to be applied which allow for examination of the reaction kinetics of single immobilized enzyme beads. In the follllowing it is shown that microfluorometry of the turnover of fluorogenic substrates may be a suitable direct method.
