Abstract

Our group has previously published work wherein microparticles with bound oxygen-sensitive and oxygen-insensitive lumiphores proved an accurate, precise and reliable tool for the quantification of localized oxygen partial pressure in vitro. Calibration between the luminescence of oxygen-sensitive lumiphore and local oxygen partial pressure allows for oxygen quantification while the luminescence of oxygen-insensitive lumiphore allows for corrections based on particle concentration. An analogous system may prove to be an equally useful tool for in vivo measurements, if certain design features are altered to address concerns such as tissue optical absorptivity and possible toxicity. Current studies focus on the design of a surface functionalized nanosphere-system as a possible approach. This work focuses on the development of this sensing technology as well as methods to allow for precise and flexible synthesis, characterization of key properties (e.g., oxygen sensing in whole blood) and optimization for in vivo conditions.
