Abstract

The use of the term anticholinergics by definition implies that this class of drugs acts primarily on cholinergic synapses, which, on the basis of our understanding of the parasympathetic nervous system, can be either muscarinic or nicotinic. Since most of the so-called anticholinergic drugs employed by neuropharmacologists and other investigators are primarily antimuscarinic, the term has generally referred to antagonists of muscarinic sites, particularly those in the parasympathetic nervous system. Although the term theoretically includes antinicotinic drugs, such as hexamethonium and other bis-quaternary amines, the discussion in this chapter will be restricted to antimuscarinic agents. Bis-quaternary agents evidently do not permeate the blood-brain barrier; however, when they are injected intraventricularly into specific brain regions, they produce severe prolonged seizures and other neurologic symptoms (Feldberg, 1963).
