Monitoring of Cell Activity: Online Oxygen Uptake Rates in Pulsed Aerated Cell Culture

Abstract

Oxygen uptake rates of adherently growing mammalian Madin Darby canine kidney cells were determined online based on the decrease of dissolved oxygen in a pulsed aerated 5 L stirred tank bioreactor. From these oxygen uptake rates and the average specific oxygen demand of the cells, viable cell numbers were estimated. The calculated cell numbers correlated well with experimentally determined viable cell numbers during exponential growth. However, differences between estimated and experimental cell numbers were identified during density dependent growth inhibition before confluence and during virus replication. Lower specific oxygen uptake rate of cell cycle arrested cells resulted in inhomogeneous cell populations. During virus replication the estimated cell numbers correlated more with the subpopulation of uninfected cells then with the experimentally determined ‘viable’ cell numbers using trypan blue dye.
