Effects of Pluronic F68 on t-PA Producing Chinese Hamster Ovary (CHO) Cells Under Agitating and Static Conditions

Abstract

Effects of agitation and addition of synthetic polymer surfactants, Pluronic F68, to culture medium on cell proliferation and metabolism of t-PA (Tissue plasminogen activator) producing recombinant Chinese Hamster Ovary (CHO) cells in serum-free medium were investigated. Increasing agitation speed reduced cell proliferation even at moderate speed (below 200 rpm), but enhanced metabolic activity. Addition of Pluronic F68 caused high cell growth rate but reduced metabolic activity including t-PA productivity not only under agitating conditions but also under static culture conditions. Generally animal cells under more unfavorable cell culture conditions have higher metabolic activity (including product productivity); thus supposedly addition of Pluronic F68 to cell culture medium could reduce the effects of fluid mechanical stresses under not only agitating condition but also static condition.
