Definition

X-linked inhibitor of apoptosis, XIAP, is a protein that contains three baculovirus inhibitors of apoptosis protein repeat (BIR) domains as well as a UBA ubiquitin-binding domain and a really interesting new gene (RING) domain. XIAP functions to prevent apoptosis by binding to caspase-3, caspase-7, and caspase-9 to inhibit caspase activity.

Structure

The XIAP protein is 497 amino acids in length and migrates on denaturing polyacrylamide electrophoresis as a ~55 kDa band. XIAP contains BIR1, BIR2, and BIR3 domains which are the defining feature IAP proteins. Each BIR domain is a zinc-binding region of about 70 amino acids. XIAP also contains a ubiquitin-binding UBA domain. Finally, the C-terminus of XIAP contains a RING domain with E3 ubiquitin ligase activity. A schematic representation of XIAP is shown in Fig. 1.
