Abstract

Current theories relating to the genesis of atherosclerosis assign an important role to smooth-muscle cells. The first stages of this disease consist of a thickening of the intima caused by the proliferation of smooth-muscle cells which have migrated from the media towards the intima [60,61]. According to certain investigators, this stimulation of the growth of smooth-muscle cells, which are usually quiescent in normal arteries, could be explained either by a mutation conferring a selective advantage on a single cell which could then divide upon stimulation [2] or by injury to the intimal barrier causing the smooth-muscle cells to be exposed to circulating serum factors [60], and, in particular, to platelet-derived growth factors which seems to be the principal mitogenic agent for smooth-muscle cells [6,62].
