Influenza Specific Cytotoxic T-Cell Clones and Immune Interferon

Abstract

We have examined several aspects of IFN- secretion by murine influenza A virus specific cytotoxic T-cell (Tc) clones. IFN- secretion is H-2 restricted and depends on contact of Tc with antigen (influenza A infected cells). There is an inverse relationship between IFN production and rapid cellular proliferation of Tc but T-cell mitogens induce IFN at all stages of cell growth. Treatment of Tc with recombinant IFN- had no effect on cytolytic activity of cloned Tc or a Tc line. Nor could we find any effect on generation of influenza specific Tc when recombinant IFN- is added during the in vitro induction of Tc. Thus we have no evidence that IFN- affects Tc proliferation or maturation. The influenza system does not lend itself to test IFN- effects on target cells, since it inhibits influenza virus expression on the cell surface of IFN susceptible cells.
