Abstract

The ability to quantitatively as well as qualitatively determine amino acid residues from protein/peptide sequencing would provide an important tool for the study of proteins, which up to recently has not been utilized.1 Edman clearly noted this; and was surprised so little emphasis was placed on the quantitative aspects of sequencing when so much additional information could be obtained by its realization.2 The method to be described attempts to fulfill this intention by having a technical ease which facilitates reproducibility and by incorporating a simple, sensitive, accurate system for quantitation; thus not only protein chemists but anyone dealing with proteins could avail themselves of the potential of quantitative sequencing. In addition the method has the flexibility to effectively treat a broad scope of sequencing problems such as rapidly deblocking and sequencing pyroglutamyl polypeptides and preventing material losses during extractive procedures (without special carriers).
