Use of Concanavalin A to Monitor Changes in Glycoprotein Synthesis during Early Mouse Development

Abstract

One- and two-dimensional gel electrophoresis have been used to examine proteins synthesized by preimplantation mouse embryos and, for each stage analyzed, about 400–600 distinct polypeptides have been resolved (Epstein and Smith, 1974, Van Blerkom and Brockway, 1975, Levinson et al., 1978). In an attempt to characterize further these proteins, we have used concanavalin A (Con A), a plant lectin that binds to mannose residues, to precipitate both total and cell surface glycoproteins synthesized during the preimplantation period. The precipitates were subsequently analyzed by two-dimensional gel electrophoresis and autoradiography.
