MR60/ERGIC-53, a Mannose-Specific Shuttling Intracellular Membrane Lectin

Abstract

Monocyte-derived macrophages express a mannoside-specific membrane lectin (Man-R) at their surface, which efficiently binds and internalizes mannosylated proteins as well as mannans (for reviews, see Stahl et al. 1984, Stahl 1992, and Taylor, this Vol.); conversely, monocytes and their precursors do not express this lectin. Knowing that the majority (80%) of the Man-R is located inside the cells and that its surface expression occurs during the first few hours of differentiation into macrophages, coming from an intracellular precursor protein pool (Lennartz et al. 1987), we decided to look for the presence of an intracellular precursor in two immature cell lines, the human promyelocytic cells HL60 and the human promonocytic cells U937, by using fluorescent neoglycoproteins.
