Peripheral Mediators of Pain

The Pharmacology of Pain

Part of the book series: Handbook of Experimental Pharmacology ((HEP,volume 130))

Abstract

Pain is a cardinal feature of normal physiological protective mechanisms to avoid tissue damage. In the periphery pain is signalled by fine C and Aδ afferent nerve fibres which respond to noxious stimuli (mechanical, heat, cold and chemical). Indeed all tissues, with the exception of the neuropil of the CNS, are innervated by such afferent fibres. However, pain is not a uniform sensation, and both the quality of pain and the initiation of protective responses are determined by many factors within the spinal cord and in higher brain structures involved in the integration and modification of nociceptive signals.
