Cosmid Cloning of Nitrogenase Structural Genes of Azospirillum Lipoferum

Summary

Cosmid gene libraries were established from two strains of Azospirillum lipoferum (ATCC 29708 and 29731). The clones carrying the nif structural genes (nifHDK) were isolated. These genes were found to be clustered and organized in the same order as in Klebsiella pneumoniae (nifHDK). After introduction into the respective K. pneumoniae nif - mutants the cloned A. lipoferum nifHDK (Ali nifHDK) restored nitrogenase activity up to approximately 2% of wildtype.
