Transmission Electron Microscopy of Cytological Specimens

Abstract

Fine needle aspiration is a rapid and convenient way of obtaining tumour cells for both cytological smears and electron microscopy. Specimens from fine needle aspirates have to be immediately fixed, for example in a cacodylate-buffered mixture of 1% glutaraldehyde and 4% formaldehyde (McDowell and Trump 1976), postfixed in buffered 1% osmium tetroxide and dehydrated in graded ethanol and propylene oxide. After infiltration in an Epon/Araldite mixture the blocks are polymerized at 60°–80°C. Semithin sections are cut with glass knives, stained with toluidine blue and used for light microscopical orientation. Ultrathin sections are cut with diamond knives, contrasted with uranyl acetate and lead citrate and examined under the electron microscope.
