Zusammenfassung
Ein zweidimensionales lichtmikroskopisches Bild eines dreidimensionalen Objekts hat einen geringen Informationsinhalt und ist aufgrund der defokussierten Projektion des gesamten Objekts in die Fokusebene mit systematischen Fehlern behaftet. Durch die Aufnahme von Fokusserien können dreidimensionale Bilder erzeugt werden. Mit der Kenntnis der dreidimensionalen optischen Übertragungsfunktion kann daraus innerhalb bestimmter Grenzen das dreidimensionale Objekt rekonstruiert werden, wie am Beispiel von Zellkernbildern gezeigt wird.
