Abstract
We have previously demonstrated the presence of an HLA class I-like molecule on human extravillous trophoblast and on a choriocarcinoma cell line, BeWo, neither of which appear to express HLA class I A, B, or C (1). In this study, we describe the characterisation of an HLA class I molecule at the nucleotide level, in the BeWo cell line. Three class I DNA probes were used in a Northern blot analysis to detect mRNA from class I or related genes. We found hybridization with all three probes, the strongest being seen with the pC800 probe (2), which corresponds to an 800bp section of the Cw3 gene, containing exons 7 and 8. We have made a cDNA library and screened it for positive clones using the p44/2 probe (3). The sequences which we have obtained are clearly derived from a single class I gene, since they are identical to each other and show a high level of homology to published class I sequences. The sequence is C-locus related, but is not identical to Cwl, Cw2, or Cw3.
