Gene expression and structure of globin genes of Carp

Summary

We cloned eight α- and nine β-globin genes in carp, with the basic structure of vertebrate globin genes, i.e., three exons interrupted with the two introns. In carp globin gene loci, the head-to-head arrangement suggests that the original directions of duplicated ancestral globin genes, prior to the α- and β-globin gene divergence may have occurred in the head-to-head direction. Carp globin genes are classified into 3 types (type I, type II, and type III) according to those nucleotide sequence identities of protein coding regions.

An RT-PCR analysis was performed to investigate the expression pattern of these three different type globin genes during development. This result indicates that carp α-and β-globin type I are embryo and adult type, and type II and III are the larval stage expressed genes.

Furthermore, we obtained a BAC clone having 170-kb insert coding type I and III globin genes. It was suggested that a 9-kb of type I cluster link to a 20 kb of type III cluster with 10 kb insertion from the restriction enzyme mapping of this BAC clone. We are currently investigating the location of each globin gene in the cluster and locus control region (LCR), which regulates the expression of globin switching.
