Antibiotic Photoaffinity Labeling Probes of Escherichia Coli Ribosomal Structure and Function

Abstract

The use of antibiotic photoaffinity label probes to identify functionally important E. coli ribosomal components is presented, emphasizing in particular recent results obtained in the authors’ laboratory with the antibiotics puromycin, chloramphenicol, and tetracycline. These results have led to the identification of 50S proteins and bases within 23S rRNA at the peptidyl transfer center as well as of proteins involved in tRNA binding to the 30S subunit. Particular stress is placed on the methodologies used in component identification and on the different approaches that are available for demonstrating the functional significance of photoaffinity labeling results.
