Effects of Iron Overload on Bone Remodelling in Pigs

Abstract

Although an iron overload in human pathology may be associated with osteoporosis, it cannot be inferred that iron plays a role in bone changes. Osteoporosis in primary hemochromatosis may be related to multi-endocrine deficiency (De60, Ro79) and in Bantu hemochromatosis to vitamin C deficiency (Wa71). Furthermore, in both cases, hepatic failure frequently associated with bone loss, may also induce osteoporosis (Lo78). In thalassemia major with secondary hemochromatosis, impaired bone mineralization and apposition associated with iron bone overload have been observed (Ve82). The role of iron in such disorders, however, has not been demonstrated. At the present time, an iron overload is also frequently observed in hemodialized patients (Go79) and; it is relevant to know whether an iron overload may play a role in renal osteodystrophy. We have therefore induced an iron overload in pigs of the same magnitude as that observed during human hemochromatosis. Bone remodelling after a 36-day iron overload was studied in growing pigs after a double tetracycline labelling (Fr69) in order to determine whether this treatment leads to bone changes.
