Norzoanthamine Suppresses PTH-Stimulated IL-6 Production in Vitro and Bone Resorption in Vivo

Abstract

Interleukin 6 (IL-6) is known as a mediator of bone résorption caused by estrogen deficiency[1]. IL-6 is produced by osteoblasts and involved in bone metabolism through osteoclast formation. Ovariectomy causes an increase of DL-6 level in bone marrow and loss of bone weight in normal mice, however ovariectomy in IL-6 defficient mice was reported not to cause loss of bone weight [2, 3]. Therefore, inhibition of IL-6 production or IL-6 activity is expected to suppress bone weight loss under the estrogen defficient condition. In vitro experimental system, EL-6 production is enhanced by PTH (parathyroid hormon) and suppressed by estrogen[4]. To find a novel osteoporotic drug, we searched for compounds with a suppressive effect on PTH-stimulated IL-6 production in MC3T3-E1, pre-osteoblastic cell line.
