Abstract

In microscopic scanning of isolated objects of size less than 1 or 2 microns for the purpose of their size determination, the effect of diffraction of light due to the finite aperture of the imaging system must be taken into account, even though the appearance of the boundary in the image is apparently distinct. For this, the theoretical equations giving the light intensity distribution in the image, for particles of varying degrees of contrast with respect to the surround, as also for different kinds of illumination of the particles, have to be used.
