Abstract

Molecular weight (mw) and number of polypeptides in the coat protein is one of the criteria used for the identification of virus species. The determination of molecular weight of the viral coat protein involves denaturing of protein subunits in the presence of sodium dodecyl sulphate (SDS) by heating in boiling water. The denatured protein is further run on an SDS-poly acrylamide gel electrophoresis (SDS-PAGE) along with protein markers of known molecular weight. The molecular weight of the unknown virus protein is estimated by comparing its mobility in the gel, with that of other proteins of known molecular weight.
