Abstract
The following protocol is one of the longest-established methods of DNA extraction and works well with a wide range of solid tissues. Proteins are digested with proteinase K and extracted with phenol chloroform. DNA is then precipitated with ethanol. The resultant DNA (10–50 μg) is of high molecular weight and is a suitable template for long polymerase chain reaction (PCR).
